Oral Wegovy®
Wegovy® tablets are a brand-name oral semaglutide, a GLP-1 receptor agonist manufactured by Novo Nordisk. It works by mimicking a hormone your body naturally produces in response to food, reducing appetite signaling and slowing gastric emptying. It is FDA-approved as a once-daily tablet for chronic weight management in adults.

Take one Wegovy® tablet once daily in the morning on an empty stomach. Swallow the tablet whole with water only (up to 4 ounces). Do not crush, split, chew, or dissolve the tablet. Wait at least 30 minutes after taking before eating any food, drinking any beverages, or taking other oral medications. If you miss a dose, skip it and take your next dose the following day. Your physician will start you at a lower dose and gradually increase it approximately every 30 days
WEGOVY® tablets may cause side effects, especially stomach-related symptoms during dose escalation. Common side effects can include nausea, diarrhea, vomiting, constipation, abdominal pain, headache, tiredness, upset stomach, dizziness, bloating, belching, gas, stomach flu, heartburn, changes in skin sensation, low blood sugar in people with type 2 diabetes, and hair loss.
Some side effects can be serious. Contact your healthcare provider right away if you develop severe or persistent stomach pain, pain that spreads to your back, vomiting that will not stop, signs of gallbladder problems, signs of dehydration, symptoms of low blood sugar, vision changes if you have diabetes, a racing heartbeat while at rest, or symptoms of an allergic reaction. WEGOVY® tablets are not recommended for patients with severe gastroparesis and should not be used by patients with a personal or family history of medullary thyroid carcinoma or Multiple Endocrine Neoplasia syndrome type 2.

What is this?
This Important Safety Information summarizes key warnings, precautions, contraindications, and possible side effects for WEGOVY® tablets (semaglutide), a prescription GLP-1 receptor agonist taken by mouth.
WEGOVY® tablets are indicated in combination with a reduced calorie diet and increased physical activity to reduce the risk of major adverse cardiovascular events in adults with established cardiovascular disease and either obesity or overweight, and to reduce excess body weight and maintain weight reduction long term in adults with obesity or in adults with overweight in the presence of at least one weight-related comorbid condition.

WEGOVY® contains semaglutide and should not be used with other semaglutide-containing products or with any other GLP-1 receptor agonist medication.
Boxed Warning: Risk of Thyroid C-Cell Tumors
In rodents, semaglutide caused thyroid C-cell tumors at clinically relevant exposures. It is unknown whether WEGOVY® causes thyroid C-cell tumors, including medullary thyroid carcinoma, or MTC, in humans.
WEGOVY® is contraindicated in patients with a personal or family history of MTC or in patients with Multiple Endocrine Neoplasia syndrome type 2, also called MEN 2.
Tell your healthcare provider if you develop symptoms of a thyroid tumor, including a lump or swelling in the neck, trouble swallowing, trouble breathing, or persistent hoarseness.
Who should not take WEGOVY® tablets?
Do not take WEGOVY® tablets if you or any of your family have ever had medullary thyroid carcinoma, or if you have Multiple Endocrine Neoplasia syndrome type 2.
Do not take WEGOVY® tablets if you have had a serious allergic reaction to semaglutide or any of the ingredients in WEGOVY® tablets.
Before taking WEGOVY® tablets
Before taking WEGOVY® tablets, tell your healthcare provider if you have or have had problems with your pancreas or kidneys, type 2 diabetes, diabetic retinopathy, stomach or digestion problems, severe gastroparesis, gallbladder problems, or if you are scheduled to have surgery or another procedure that uses anesthesia or deep sedation.
Tell your healthcare provider if you are pregnant, planning to become pregnant, breastfeeding, or planning to breastfeed.
Tell your healthcare provider about all medicines you take, including prescription medicines, over-the-counter medicines, vitamins, and herbal supplements. WEGOVY® may affect the way some medicines work, and some medicines may affect the way WEGOVY® works.
How to take WEGOVY® tablets
Take WEGOVY® tablets exactly as your healthcare provider tells you to take them.
Take one WEGOVY® tablet by mouth once daily on an empty stomach in the morning with water, using no more than 4 ounces of water.
Do not take WEGOVY® tablets with liquids other than water.
Do not take more than one WEGOVY® tablet each day.
Swallow WEGOVY® tablets whole. Do not split, crush, chew, or dissolve the tablet.
After taking a WEGOVY® tablet, wait at least 30 minutes before eating food, drinking beverages, or taking other oral medications.
Use WEGOVY® tablets with a reduced calorie diet and increased physical activity.
Dosage and dose escalation
The recommended starting dosage of WEGOVY® tablets is 1.5 mg taken orally once daily for 30 days.
The recommended dose escalation schedule is 1.5 mg once daily on days 1 through 30, 4 mg once daily on days 31 through 60, 9 mg once daily on days 61 through 90, and 25 mg once daily on day 91 and onward.
The recommended maintenance dosage of WEGOVY® tablets is 25 mg orally once daily.
If you do not tolerate a dose during dose escalation, your healthcare provider may consider delaying dose escalation.
Acute pancreatitis
Acute pancreatitis, including fatal and non-fatal hemorrhagic or necrotizing pancreatitis, has been observed in patients treated with GLP-1 receptor agonists, including WEGOVY®.
Stop taking WEGOVY® and contact your healthcare provider right away if you have severe pain in your stomach area that will not go away, with or without nausea or vomiting. The pain may be felt from your abdomen to your back.
Gallbladder problems
WEGOVY® is associated with an increased occurrence of gallbladder problems, including gallstones and inflammation of the gallbladder.
Contact your healthcare provider if you develop symptoms of gallbladder problems, including pain in the upper stomach area, fever, yellowing of the skin or eyes, or clay-colored stools.
Low blood sugar
WEGOVY® can lower blood glucose and may increase the risk of low blood sugar, especially in patients with type 2 diabetes who also take insulin or an insulin secretagogue such as a sulfonylurea.
Tell your healthcare provider if you take medicines to treat diabetes. Your diabetes medication dose may need to be adjusted to reduce the risk of low blood sugar.
Symptoms of low blood sugar may include dizziness, sweating, confusion, shakiness, weakness, headache, fast heartbeat, hunger, irritability, or feeling jittery.
Kidney problems due to dehydration
WEGOVY® may cause nausea, vomiting, diarrhea, or other symptoms that can lead to dehydration. Dehydration may cause kidney problems, including acute kidney injury.
Your healthcare provider may monitor kidney function if you report side effects that could lead to dehydration, especially when starting treatment or increasing the dose.
Contact your healthcare provider if you have vomiting, diarrhea, inability to keep fluids down, reduced urination, dizziness, or symptoms of dehydration.
Severe gastrointestinal side effects
WEGOVY® has been associated with gastrointestinal side effects, sometimes severe.
WEGOVY® is not recommended in patients with severe gastroparesis.
Contact your healthcare provider if you experience severe, persistent, or worsening nausea, vomiting, diarrhea, constipation, stomach pain, bloating, or symptoms that interfere with hydration or normal eating.
Allergic reactions
Serious allergic reactions, including anaphylaxis and angioedema, have been reported with WEGOVY®.
Stop taking WEGOVY® and seek medical help right away if you develop symptoms of a serious allergic reaction, including swelling of the face, lips, tongue, or throat; trouble breathing or swallowing; severe rash or itching; fainting; dizziness; or very rapid heartbeat.
Diabetic retinopathy complications
Diabetic retinopathy complications have been reported in trials with semaglutide. Rapid improvement in glucose control has been associated with temporary worsening of diabetic retinopathy.
Patients with type 2 diabetes and a history of diabetic retinopathy should be monitored for progression of diabetic retinopathy.
Contact your healthcare provider if you experience changes in vision while taking WEGOVY® tablets.
Heart rate increase
WEGOVY® has been associated with increases in resting heart rate.
Your healthcare provider may monitor your heart rate during treatment. Tell your healthcare provider if you experience palpitations or a racing heartbeat while at rest.
If you experience a sustained increase in resting heart rate, WEGOVY® may need to be discontinued.
Pulmonary aspiration during anesthesia or deep sedation
WEGOVY® delays gastric emptying. Rare postmarketing reports of pulmonary aspiration have occurred in patients receiving GLP-1 receptor agonists during elective surgeries or procedures requiring general anesthesia or deep sedation.
Tell your healthcare providers that you take WEGOVY® tablets before any planned surgery or procedure involving anesthesia or deep sedation.
Possible side effects
The most common side effects of WEGOVY® include nausea, diarrhea, vomiting, constipation, abdominal pain, changes in skin sensations, headache, tiredness or fatigue, upset stomach, dizziness, bloating, belching, low blood sugar in people with type 2 diabetes, gas, stomach flu, heartburn, and hair loss.
Other reported side effects include acute pancreatitis, gallbladder problems, acute kidney injury, increased heart rate, low blood pressure, fainting, dysgeusia or altered taste, increases in amylase and lipase, rash, hives, angioedema, anaphylaxis, ileus, intestinal obstruction, severe constipation, and fecal impaction.
Talk to your healthcare provider about any side effect that bothers you or does not go away.
Drug interactions
Tell your healthcare provider about all medications you take, including prescription medications, over-the-counter medications, vitamins, and herbal supplements.
WEGOVY® may increase the risk of low blood sugar when used with insulin or an insulin secretagogue, such as a sulfonylurea.
WEGOVY® delays gastric emptying and may affect the absorption of oral medications. Your healthcare provider may consider additional clinical or laboratory monitoring when WEGOVY® is taken with oral medications that have a narrow therapeutic index or require clinical monitoring.
Pregnancy
WEGOVY® may cause fetal harm. For patients receiving WEGOVY® for cardiovascular risk reduction or weight reduction, discontinue WEGOVY® when pregnancy is recognized.
Patients planning pregnancy should stop WEGOVY® at least 2 months before a planned pregnancy because of the long half-life of semaglutide.
There is a pregnancy exposure registry for women who use WEGOVY® during pregnancy. Talk to your healthcare provider about how to take part in the registry.
Breastfeeding
Breastfeeding is not recommended during treatment with WEGOVY® tablets.
Use in pediatric or geriatric patients
It is not known if WEGOVY® tablets are safe and effective for use in people under 18 years of age.
Older adult patients may be more sensitive to side effects based on individual health status, medical history, and other medications. Your healthcare provider will determine whether WEGOVY® tablets are appropriate for you.
Type 2 diabetes
In patients with diabetes, blood glucose should be monitored before starting WEGOVY® and during treatment.
If you take insulin or a sulfonylurea, your healthcare provider may reduce the dose to lower the risk of hypoglycemia.
Missed Dose
If a dose of WEGOVY® tablets is missed, skip the missed dose and take the next dose the following day.
Do not take more than one WEGOVY® tablet in a day to make up for a missed dose.
Overdose
If you take too much WEGOVY®, you may have severe nausea, severe vomiting, and severe low blood sugar.
In case of overdose or accidental exposure, call your healthcare provider, contact Poison Control at 1-800-222-1222, or go to the nearest hospital emergency room right away.
Storage
Store WEGOVY® tablets at room temperature between 20°C to 25°C / 68°F to 77°F, with excursions permitted between 15°C to 30°C / 59°F to 86°F.
Store and dispense WEGOVY® tablets in the original bottle.
Store WEGOVY® tablets in the original closed bottle until you are ready to take one. Do not store the tablets in another container.
Store WEGOVY® tablets in a dry place away from moisture.
Keep WEGOVY® and all medicines out of the reach of children.
When to contact your provider
Contact your healthcare provider if you experience persistent or severe nausea, vomiting, diarrhea, constipation, abdominal pain, bloating, dehydration, reduced urination, symptoms of low blood sugar, vision changes, racing heartbeat while at rest, symptoms of gallbladder disease, symptoms of pancreatitis, or any side effect that bothers you or does not go away.
Seek emergency medical care
Seek emergency medical care if you experience severe allergic reaction, swelling of the face or throat, trouble breathing, fainting, severe dehydration, severe abdominal pain that does not go away, severe vomiting, symptoms of pancreatitis, symptoms of severe low blood sugar, or any sudden or severe symptoms after taking WEGOVY® tablets.

Tell your healthcare provider if you experience side effects or symptoms that concern you.
